چکیده

Confocal theta fluorescence microscopy using two-photon absorption and annular apertures. In a confocal theta fluorescence microscope, a second objective lens placed orthogonally to the illumination objective lens is used for detection. The addition of annular apertures to such an arrangement leads to a reduction of the volume at half maximum of the point spread function. We show that when the sample is illuminated in a two-photon absorption mode a threedimensional resolution improvement by more than 60% can be achieved in the limit of a narrow annular aperture, The volume at half maximum of a confocal theta microscope with a numerical aperture of 0.75 and an illumination wavelength of 800 nm is then reduced from 12.3 .to 4.6 attoliters. An important advantage of the two-photon absorption mode is that no diffraction side lobes are present when an annular illumination aperture is used.

Entangled-Photon Microscopy

All forms of microscopy that have previously been developed rely on classical sources of light, which are relatively noisy by virtue of the photons arriving randomly in time and position. We propose a new technique of two-beam optical microscopy using a nonclassical light source comprising entangled photon pairs generated by the process of nonlinear optical parametric downconversion. Entangled-...

Integrated photoacoustic, confocal, and two-photon microscope.

The invention of green fluorescent protein and other molecular fluorescent probes has promoted applications of confocal and two-photon fluorescence microscopy in biology and medicine. However, exogenous fluorescence contrast agents may affect cellular structure and function, and fluorescence microscopy cannot image nonfluorescent chromophores. We overcome this limitation by integrating optical-...

An evaluation of two-photon excitation versus confocal and digital deconvolution fluorescence microscopy imaging in Xenopus morphogenesis.

The ability to visualize cell motility occurring deep in the context of opaque tissues will allow many currently intractable issues in developmental biology and organogenesis to be addressed. In this study, we compare two-photon excitation with laser scanning confocal and conventional digital deconvolution fluorescence microscopy, using the same optical configuration, for their ability to resol...

Fundamental improvement of resolution with a 4Pi-confocal fluorescence microscope using two-photon excitation

A new type of confocal fluorescence microscope with an unprecedented resolution is proposed. Calculations demonstrate that an axial resolution on the order of 100 nm can be achieved in a confocal microscope with enlarged aperture when two-photon excitation is applied. This axial resolution is up to four times higher than that ofa confocal fluorescence microscope. The proposed microscope yields ...

When Two Is Better Than One: Elements of Intravital Microscopy

O ver the last 20 years, many cell biological studies have moved from the single-cell level to the tissue level, and even to whole animals. This progress has been led by developments in fl uorescence microscopy that permit molecular observations from single cells within intact tissue or animals. Concurrent developments in fl uorescent probes, especially the cloning of the Green Fluorescent Prot...
